Not a technical question really... 

 

I'm looking for a div to just bounce once when rolled over, can you use an ease that starts and ends at the same position but has a bounce ease attached to it?

 

Or do I need to tween it to a position, and then tween it back?

No, a tween must have different starting and ending values in order to run.

 

You could create a TweenMax tween with yoyo:true, repeat:1 which means it will play forwards to your end value and then rewind to the start values.

 

Not really sure what kind of bounce you need, but here is a very simple example of a scale bounce that uses 2 tweens back to back:
